A01 - Systematic dissection of cytosolic DNA sensing and signalling with nanobodies and proteomics
Cytosolic DNA sensors have evolved to counteract pathogen infections and cell damage. The regulation and coordination of the various signaling pathways and effector mechanisms is complex and involves intra-cellular trafficking of signaling components and as well as other mechanisms. We will apply cutting-edge technology to dissect the molecular regulation of AIM2 and cGAS-STING based signalling and effectors: we will apply spatial mass spectrometry-based proteomics to assess trafficking of signaling components at a systems level; we will further use nanobodies as tools to precisely perturb assembly, post-translational modifications, as well as trafficking of signaling components on a molecular level.
