A28 - Cell-fate decision upon RNA sensing by RIG-Iike-I receptors and viral restriction factors
We have recently demonstrated that RIG-I and OAS1 share ligand requirements regarding short dsRNAs with a 5’-triphosphate moiety. Both receptor systems influence the cell fate with RIG-I activation leading to type I IFN secretion and induction of an antiviral cellular state and OAS1/RNase L inhibiting viral replication via translational inhibition and ultimately cellular demise. We will decipher the molecular activation requirements of OAS1 and analyse how the intricate interplay of downstream events upon RIG-I and OAS1 and other viral restriction factors shape the cell fate and immunogenicity of the RNA sensing event in order to induce an effective antiviral immune response.
